Abstract

Abstract: Moringa (Moringa oleifera Lam.) is a medicinal plant that has many health benefits and has been widely used as herbal medicine for certain diseases. To make an ingredient into a standardized test is needed because the chemical composition of a plant is not always the same. Moringa growth in South Sumatra is spread with varying altitudes, ranging from lowlands, medium, and high. The altitude is one of the factors that affect the composition of the metabolite compounds in Moringa leaves. The purpose of this study was to determine the metabolite profile and the quantity of moringa leaf metabolites from different heights of growth using a non-targeted metabolomic analysis approach using the GC-MS instrument. Sampling locations were carried out using a stratified purposive sampling method based on altitude, namely from Tebing Gerinting Village, Ogan Ilir District (35 masl), Masam Bulau Village, Lahat District (570 masl), and Bangun Rejo Village, Pagar Alam District (795 masl) South Sumatra Province. The results obtained were marked by differences in the types and metabolites detected. Moringa leaves in Bangun Rejo Village (795 masl) and Masam Bulau Village (570 masl) showed 29 types of metabolites with a total type of compound of 98.9% and 94.34%, respectively, while Tebing Gerinting Village, Ogan Ilir (35 masl) showed 27 types of metabolites with a total of 83.52%. Moringa leaves growing in Tebing Gerinting Village (35 masl) showed the dominant metabolite compounds Palmitic acid, methyl ester (with a percent area of 12.39%) and Linolenic acid, methyl ester (32.93%), Masam Buluu Village (570 masl) with Linolenic aci, methyl ester (14.69%) and Oleic acid, methyl ester (17.28%), and Bangun Rejo Village (795 masl) with Palmitic acid methyl ester (22.78%) and Oleic acid, methyl ester ( 37.82%). PCA analysis, Moringa leaves from Tebing Gerinting Village (35 masl) and Masam Bulau Village (570 masl) showed that 5 metabolites detected were the same and different. Moringa leaves from Masam Buluu Village (570 masl) and Bangun Rejo Village (795 masl) showed 2 types of the same metabolite compounds with various variations. The difference in the profile of the metabolites and metabolites of Moringa leaves is possible due to differences in altitude, air temperature, soil temperature, and light intensity at each location where the Moringa plant grows. Keyword: GC-MS, Altitude, Moringa oleifera, PCA, Metabolite Profile Abstrak: Kelor (Moringa oleifera Lam.) merupakan tanaman obat yang mempunyai banyak khasiat bagi kesehatan dan telah banyak digunakan sebagai obat herbal untuk penyakit tertentu. Untuk menjadikan suatu bahan menjadi obat herbal diperlukan standarisasi pengujian, karena komposisi kimia suatu tanaman tidak selalu sama. Pertumbuhan kelor di Sumatera Selatan tersebar dengan ketinggian tempat yang bervariasi, mulai dari dataran rendah, sedang dan tinggi. Ketinggian tempat tumbuh diduga menjadi salah satu faktor yang mempengaruhi komposisi senyawa metabolit daun kelor. Tujuan dari penelitian ini yaitu, untuk mengetahui profil metabolit dan kuantitas kelimpahan senyawa metabolit daun kelor dari ketinggian tempat tumbuh yang berbeda dengan pendekatan analisis metabolomik non-target menggunakan instrument GC-MS. Penentuan lokasi pengambilan sampel dilakukan dengan metode stratified purposive sampling berdasarkan ketinggian, yaitu dari Desa Tebing Gerinting Kab. Ogan Ilir (35 mdpl), Desa Masam Bulau Kab. Lahat (570 mdpl), dan Desa Bangun Rejo Kab. Pagar Alam (795 mdpl) Provinsi Sumatera Selatan. Diperoleh hasil yang ditandai dengan perbedaan jenis dan kelimpahan senyawa metabolit yang terdeteksi. Daun kelor yang tumbuh di Desa Bangun Rejo (795 mdpl) dan Desa Masam Bulau (570 mdpl) menunjukkan 29 jenis senyawa metabolit dengan total kelimpahan jenis senyawa masing-masing sebesar 98,9% dan 94,34%, sedangkan Desa Tebing Gerinting, Ogan Ilir (35 mdpl) menunjukkan 27 jenis senyawa metabolit dengan total kelimpahan sebesar 83,52%. Daun kelor yang tumbuh di Desa Tebing Gerinting (35 mdpl) menunjukkan senyawa metabolit dominan Palmitic acid methyl ester (dengan persen area sebesar 12,39%) dan Linolenic acid methyl ester (32,93%), Desa Masam Bulau (570 mdpl) dengan senyawa Linolenic acid methyl ester (14,69%) dan Oleic acid methyl ester (17,28%), serta Desa Bangun Rejo (795 mdpl) dengan senyawa Palmitic acid methyl ester (22,78%) dan Oleic acid, methyl ester (37,82%). Berdasarkan analisis PCA, daun kelor dari Desa Tebing Gerinting (35 mdpl) dan Desa Masam Bulau (570 mdpl) menunjukkan 5 senyawa metabolit yang terdeteksi sama dengan kelimpahan yang berbeda. Daun kelor dari Desa Masam Bulau (570 mdpl) dan Desa Bangun Rejo (795 mdpl) menunjukkan 2 jenis senyawa metabolit sama dengan kelimpahan berbeda. Perbedaan profil metabolit dan kelimpahan senyawa metabolit daun kelor memungkinkan karena perbedaan ketinggian tempat, suhu udara, suhu tanah, dan intensitas cahaya setiap lokasi tumbuh tanaman kelor.
