Abstract
Dahlia tubers contain components that are efficacious as prebiotics, namely inulin. The inulins levels in dahlia tubers are quite high, but their use is still limited. Therefore, this research was conducted to develop the potential of dahlia tubers as a powder drink that is efficacious as a prebiotic. This study used 5 powder drink formulas which were then tested hedonic. Respondents most liked formula 1 which contained 15% dahlia tuber powder and 85% ginger powder. Formula 1 was tested for quality standardization based on SNI 01-4320-1996. As a result, Formula 1 meets the requirements both in terms of organoleptic (normal, according to the spices used), water content (1,5%), total ash content (1%), sugar content (3,91%), metal contamination (Cu: 0,94 mg/kg, Zn: 7,30 mg/kg, Pb < 0,0628 mg/kg, As and Sn were not detected) and microbial contamination (ALT: 3,0 x 102 colonies/gram and coliform test) < 3 APM/gram). Determination of prebiotic activity was carried out by viability test using total plate count method. Powder drink formula 1 can stimulate the growth of Lactobacillus acidophilus by 3,95 X 109 CFU/mL, lower than the test group with dahlia tuber powder samples only, which produces probiotics of 26,80 X 109 CFU/mL. The antibacterial properties of Lactobacillus acidophilus metabolites grown on media containing powdered drink formula 1 were tested by disc diffusion method and produced an inhibition zone against Eschericia coli in the sensitive category with an inhibitory diameter of 13,33 + 1,53 mm. Keywords: Dahlia tubers, Inulin, Prebiotics, Viability, Antibacterial Umbi dahlia mengandung komponen yang berkhasiat sebagai prebiotik, yakni inulin. Kadar inulin pada umbi dahlia cukup tinggi, namun pemanfaatannya masih terbatas. Oleh sebab itulah dilakukan penelitian ini untuk mengembangkan potensi umbi dahlia sebagai minuman serbuk yang berkhasiat sebagai prebiotik. Penelitian ini menggunakan 5 formula minuman serbuk yang kemudian diuji hedonik. Responden paling menyukai formula 1 yang mengandung 15% serbuk umbi dahlia dan 85% serbuk jahe. Formula 1 diuji standarisasi mutunya berdasarkan SNI 01-4320-1996. Hasilnya, Formula 1 memenuhi persyaratan baik dari segi organoleptik (normal, sesuai rempah-rempah yang digunakan), kadar air (1,5%), kadar abu total (1%), kadar gula (3,91%), cemaran logam (Cu: 0,94 mg/kg, Zn: 7,30 mg/kg, Pb <0,0628 mg/kg, As dan Sn tidak terdeteksi) dan cemaran mikroba (ALT: 3,0 x 102 koloni/gram dan uji coliform < 3 APM/gram). Penentuan aktivitas prebiotik dilakukan dengan uji viabilitas menggunakan metode total plate count. Minuman serbuk formula 1 dapat menstimulasi pertumbuhan bakteri Lactobacillus acidophilus sebesar 3,95 X 109 CFU/mL, lebih rendah dari kelompok uji dengan sampel serbuk umbi dahlia saja, yang menghasilkan bakteri probiotik sebesar 26,80 X 109 CFU/mL. Sifat antibakteri metabolit Lactobacillus acidophilus yang telah ditumbuhkan pada media mengandung minuman serbuk formula 1 diuji dengan metode difusi cakram dan menghasilkan zona hambat terhadap bakteri Eschericia coli pada kategori sensitif dengan diameter hambat 13,33 + 1,53 mm.
