Abstract

This research was conducted to determine the prebiotic activity in combination powder drink of lotus seeds and ginger. Lotus seeds are used as a prebiotics, and ginger as a flavor enhancer in powdered drinks. The research was conducted by looking at the effect of the combined powder drink on the growth of L. acidophilus. The combination powder drink formula consist of F1, F2, F3, F4, F5. Hedonic test was conducted to see the most preferred formulation. Then the combined powder drinks with the most preferred formula was tested for quality standard based on SNI 01-4320-1996. L. acidophilus viability test was performed using the total plate count method. And antibacterial test of L. acidophilus metabolites was carried out by the disc diffusion method. The results of the hedonic test showed that formula 1 was the most preferred. Formula 1 powder drink meets the quality standard of SNI 01-4320-1996. With normal organoleptic and typical spices, water content 2,5%, total ash content 1%, sugar content 3,91%, Pb content: <0,06 mg/kg, Cu: 1,6 mg/kg, Zn: 13,61 mg/kg, Sn and As not detected, ALT microbial contamination: 3x102 colonies/gr, and coliform < 3 APM/gr. The results of the viability test showed that the combination powder drink stimulated the growth of the most L. acidophilus with the number of bacteria 64,80 x109 CFU/mL. The antibacterial ability of the L. acidophilus metabolite in the combined powder drink media in inhibiting E. coli was included in the strong category with an inhibitory diameter of 14,16 ± 0,76 mm. Keyword : Powder drink, lotus seed, prebiotic, L. acidophilus Penelitian ini dilakukan untuk mengetahui aktivitas prebiotik pada minuman serbuk kombinasi biji teratai dan jahe. Biji teratai digunakan sebagai sumber prebiotik, dan jahe sebagai penguat cita rasa pada minuman serbuk. Penelitian dilakukan dengan melihat pengaruh minuman serbuk kombinasi terhadap pertumbuhan L. acidophilus. Formula minuman serbuk kombinasi terdiri dari F1, F2, F3, F4, F5. Dilakukan uji hedonik untuk melihat formulasi yang paling disukai. Kemudian minuman serbuk kombinasi dengan formula yang paling disukai diuji standar mutu berdasarkan SNI 01-4320-1996. Dilakukan uji viabilitas L. acidophilus dengan metode total plate count. Serta uji antibakteri metabolit L. acidophilus pada media minuman serbuk dilakukan dengan metode difusi cakram. Hasil uji hedonik yang didapatkan menunjukkan formula 1 yang paling disukai. Minuman serbuk formula 1 memenuhi standar mutu SNI 01-4320-1996. Dengan organoleptik yang normal dan khas rempah-rempah, kadar air 2,5%, kadar abu total 1%, kadar gula 3,91%, kadar Pb: <0,06 mg/kg, Cu: 1,6 mg/kg, Zn: 13,61 mg/kg, logam Sn dan As tidak terdeteksi, cemaran mikroba ALT: 3x102 koloni/gr, dan coliform <3 APM/gr. Adapun hasil uji viabilitas didapatkan minuman serbuk kombinasi menstimulasi pertumbuhan L. acidophilus terbanyak dengan jumlah bakteri 64,80 x109 CFU/mL. Kemampuan antibakteri metabolit L. acidophilus pada media minuman serbuk kombinasi dalam menghambat E. coli termasuk kategori kuat dengan diameter hambat 14,16 0,76 mm. Kata kunci: Minuman serbuk, biji teratai, prebiotik, L. acidophilus
