Abstract

A secondary metabolic compound was isolated fforn ethyl acetate fraction of sambiloto plant extract (Andrographis paniculata Ness). The extraction was done by maceration method. Separation and purification was counducted by chromatographic technique to obtained pure compounds of white crystal 55 mg. The structure of this compound was determined with UV, IR, NMR 1-D ('HNMR, 13C- NMR, and DEPT), 2-D (HMQC, HMBC, and COSY), and it’s antibacterial activity was tested by diffusion method using Escherichia coli and Shigella dysentriae as bacterial tested. Based on spectral data analysis could be concluded that pure compound that has been isolated was diterpenoid lactone Andrographolide (C20H30O5). The test results showed that the antibacterial activity of the extract at a concentration of 625 ppm viscous able to inhibit the growth of bacteria Escherichia coli and Shigella dysentriae inhibitory diameter 5 mm and 6 mm, whereas the pure compound has inhibitory diameter of 7 mm and 8 mm after reduction with paper discs. The pure compounds of andrographolide has more antibacterial activity than crude extract of sambiloto. The inhibition of amoxicillin with a diameter of 15 mm has more antibacterial activity than extracts and pure compounds to Shigella dysentriae and Escherichia coli than concentrations of 625 ppm.
