Abstract

Tembesu (Fagraea fragrans Roxb) has been reported to have antibacterial activity, anti inflammation, antifungal, antioxidant, antiage, anticancer, antiviral and antiflasmodial, but there have been no reported on isolating and antibacterial activity of alkaloid compound from the steam bark of this plant. Based on it, this research aimes to isolate alkaloid compound and determine the antibacterial activity of the isolated alkaloid compound from diethyl ether extract agains Staphylococcus aureus and Pseudomonas aeruginosa bacteria. The Purification process of alkaloid compound was carried out by extraction (meceration), fractination (partition), separation and purification. Meceration used diethyl ether solvent, partition with hydrochloric acid, ammonia and continued with chlorofom solvent. Separation and purification used the gravity column chromatography method. The isolated alkaloid compound was obtained as 25 mg. Stucture analysis of the isolated alkaloid compound using LCMS/MS and NMR spectroscopy ( 1H-NMR, 13C-NMR and DEPT 135), show that the isolated alkaloid compound was a pyridine alkaloid group which had similar structure to the oliveramine compound. Antibacterial activity test againts S. aureus and P. aeruginosa of alkaloid compound from the steam bark of F. fragrans resulted in a inhibition zone diameter of 0.00 mm at concentrations 1000 ppm, 500 ppm, 250 ppm and 125 ppm, compared to the positive control tetracycline (1000 ppm) resulted 10.05 mm against S. aureus bacteria and 23.6 mm against P. aeruginosa bacteria. The results of the inbhibition zone diameter data showed that the alkaloid campound isolated of F. fragrans has no antibacterial activity.
