Abstract

Avocado (Persea americana Mill.) contains phenolic compounds that have the potential to facilitate urine output. The aims of tsis study were to determined of phenolic compounds and the effect of dose variations on the diuretic effect. The animal test were divided into 6 groups consisting of the normal group was given distilled water, the positive group bewasing given furosemide 40 mg/KgBW, the negative group was given 1% NaCMC, and the treatment groups 1, 2, and 3 were given the ethyl acetate fraction of avocado leaves at a dose of 25, 50, and 100 mg/KgBW. Data were analyzed by one way ANOVA. The total phenolic content in the ethanol extract and ethyl acetate fraction is 85.43 mgGAE/g and 64.67 mgGAE/g respectively. Urine volume and levels of Na+, Cl-, and K+ ions at a dose of 100 mg/KgBW were not significantly different from the positive control group (p>0.05). The effective dose of ethyl acetate fraction of avocado leaves is 29.53 mg/KgBB. Increasing the dose indicated an increase in urine volume and ion content in the urine. Penelitian ini bertujuan untuk menentukan kadar senyawa fenolik dan mengetahui pengaruh variasi dosis terhadap efek diuretik. Hewan uji dibagi menjadi 6 kelompok yang terdiri dari kelompok normal diberi akuades, kelompok positif diberi furosemid 40 mg/KgBB, kelompok negatif diberi NaCMC 1%, dan kelompok perlakuan 1, 2, dan 3 diberi fraksi etil asetat daun alpukat dengan dosis 25, 50, dan 100 mg/KgBB. Data dianalisis dengan one way ANOVA. Kadar fenolik total pada ekstrak etanol dan fraksi etil asetat secara berurutan sebesar 85,43 mgGAE/g dan 64,67 mgGAE/g. Volume urine dan kadar ion Na+, Cl-, dan K+ dosis 100 mg/KgBB tidak berbeda signifikan terhadap kelompok kontrol positif (p>0,05). Dosis efektif fraksi etil asetat daun alpukat sebesar 29,53 mg/KgBB. Peningkatan dosis menunjukkan peningkatan volume urine dan kandungan ion dalam urine. Kata kunci : Persea americana Mill., kadar total fenolik, aktivitas diuretik, dosis efektif
