Abstract

Free radicals are relatively unstable molecules, with atoms in their outer orbits having one or more unpaired electrons (Khaira, 2010). Antioxidants are substances that are needed to protect the body from free radical compounds (Winarsi, 2007). One of the plants that has potential as a source of antioxidants is tea (Camellia sinensis L.) (Mondal et al., 2004). Extraction using the ultrasound-assisted extraction method is still focused on optimization (Sun et al., 2019). Important operational parameters for optimization include extraction time, ratio of solvent to simplicia, and ethanol concentration. The largest percentage yield was obtained from the run 11 extracts (30 minutes, 30% ethanol concentration, ratio 30) which was 38.750%. While the smallest percentage yield was 15,000 Obtained from extract run 13 (time 10 minutes, ethanol concentration 70%, ratio 10). Total flavonoid content (TFC) showed that run 4 (10 minutes time, 30% ethanol concentration, ratio 30) had the highest flavonoid content value of 64,895 %CE. While run 1 (30 minutes, 70% ethanol concentration, ratio 30) obtained the lowest value of 29.330 %CE. Phenolic content (TPC) from 15 runs, the highest value was obtained, namely run 4 for 10 minutes, 30% ethanol concentration, ratio 30 with a value of 4,530 %GAE. Meanwhile, the lowest phenolic content was run 1 (30 minutes, 70% ethanol concentration, ratio 10) with a value of 1.444 %GAE. The antioxidant activity of IC50 against 15 runs obtained the lowest value, namely at run 2 (10 minutes, concentrating 70%, ratio 30) with a value of 25.768 ppm categorized as a very strong antioxidant. While the highest IC50 value is at run 12 (30 minutes, concentrating 30%, ratio 10) with a value of 44.688 ppm. Radikal bebas adalah suatu molekul yang relatif tidak stabil, dengan atom pada orbit terluarnya memiliki satu atau lebih elektron yang tidak berpasangan (Khaira, 2010). Antioksidan merupakan substansi yang diperlukan untuk melindungi tubuh dari senyawa radikal bebas (Winarsi, 2007). Salah satu tanaman yang memiliki potensi sebagai sumber antioksidan adalah teh (Camellia sinensis L.) (Mondal dkk., 2004). Ektraksi dengan metode ultrasound-assisted extraction masih berfokus pada optimasi (Sun dkk., 2019). Paramater operasional penting untuk optimalisasi diantaranya berupa waktu ekstraksi, rasio perbandingan pelarut dengan simplisia, serta konsentrasi etanol. Persen rendemen terbesar diperoleh dari ekstrak run 11 (waktu 30 menit, konsentrasi etanol 30%, rasio 30) yakni sebesar 38,750%. Sedangkan persen rendemen terkecil yakni 15,000 diperoleh dari ekstrak run 13 (waktu 10 menit, konsentrasi etanol 70%, rasio 10). Kadar flavonoid total (TFC) menunjukan bahwa run 4 (waktu 10 menit, konsentrasi etanol 30%, rasio 30) memiliki nilai kadar flavonoid tertinggi sebesar 64,895 %CE. Sedangkan run 1 (waktu 30 menit, konsentrasi etanol 70%, rasio 30) memperoleh nilai terendah sebesar 29,330 %CE. Kadar fenolik (TPC) dari 15 run yang dilakukan diperoleh nilai kadar tertinggi yaitu run 4 kondisi waktu 10 menit, konsentrasi etanol 30%, rasio 30 dengan nilai 4,530 %GAE. Sedangkan kadar fenolik terendah yaitu run 1 (waktu 30 menit, konsentrasi etanol 70%, rasio 10) dengan nilai 1,444 %GAE. Aktivitas antioksidan IC50 terhadap 15 run diperoleh nilai terendah yaitu pada run 2 (waktu 10 menit, konsentrasi 70%, rasio 30) dengan nilai 25,768 ppm dikategorikan sebagai antioksidan sangat kuat. Sedangkan nilai IC50 tertinggi yaitu pada run 12 (waktu 30 menit, konsentrasi 30%, rasio 10) dengan nilai 44,688 ppm Kata Kunci : Radikal Bebas, Antioksidan,Teh, Ultrasound-Asissted Extraction, Persen Rendemen, TFC, TPC, IC50
