Abstract

Green tea (Camellia sinensis L.) contain flavonoid have been using antioxidant. The encapsulated green tea extract can be used as an active substance in antiaging cosmetics, one of which is peel-off gel mask preparation. In this study, green tea nano-extracts were formulated in a peel-off gel mask preparation and then the antiaging activity was determined by the in vitro method.Extraction of green tea with this ultrasonication method resulted in an extract yield of 10.26%. The characterization of tea extract included organoleptic, moisture content of 7.0%, and drying shrinkage of 8.6%. The phytochemical screening of green tea extract was positive for flavonoids, alkaloids, tannins, steroids and TLC analysis using DPPH (1,1-diphenyl-2-picrihydrazil) which showed the tea extract contained antioxidant compounds. The encapsulation process can increase antioxidant activity. The results of the encapsulation of the extract using the ionic gelation method using the Chitosan-NaTPP matrix showed that the nano-extract had a particle size of 957.5 nm with a PDI value of 0.783 and a zeta potential of 24.7 mV. The characterization of the green tea extract nano peel-off gel mask included homogeneity, organoleptic, viscosity, spreadability, adhesion, pH, drying time, and stability that fell within the range. Total flavonoid content in green tea extract was 245.78 mg/g. The extracts, nano-extracts, and gel formulas were analyzed for their antioxidant activity using DPPH and vitamin C as comparisons. The IC50 values of vitamin C, extract, nano-extract, and gel formula were 2,332 g/mL, respectively; 57.791 g/mL; 38.851 g/mL; and 103.184 g/mL. There was a significant difference (p<0.05) between vitamin C and extract, nano-extract, gel formula, wherein the peel-off nano green tea extract gel mask showed moderate antioxidant activity.
