Abstract

Antioxidant is agent that can bind free radicals and has potential as anti aging. The problem of skin aging has occurred in the community caused by free radicals. Therefore we need a cosmetic preparation that can overcome skin aging. Chlorella (Chlorella vulgaris) has a natural antioxidant activity that can be derived from secondary metabolites such as flavonoids and phenolics that can be used as anti aging. This research was started from the process extracting of chlorella powder using the Ultrasound Assisted Extraction (UAE) method with 96% ethanol solvent followed by extract characterization, analysis of phytochemical content, and thin layer chromatography (TLC), and total flavonoid extract. Furthermore, the extract was made in the form of nanoparticles with chitosan polymer and sodium tripolyphosphate (NaTPP) with a concentration 0,1% and continued with the characterization of nanoparticles. Chlorella extract nanoparticles were used as an active substance in the manufacture of peel off gel mask preparations, then an evaluation of the mask preparation was carried out and antioxidant activity through in vitro free radical scavenging using DPPH (1,1-difenil-2-pikrilhidrazil). Chlorella extract has a yield of 5,18% and contains alkaloids, flavonoids, steroids, saponins, and phenolic compounds. The results of the TLC test indicate that there are secondary metabolites that have the potential as antioxidants. The total flavonoid content of the extract contained in the extract was 67,486 mg/g. The results of the nanoparticles characterization of the extract showed that the percentage of transmittance was 87,4%, particle size of 549,5 nm, PDI (poly dispersity index) 0,375 and zeta potential +43,35 mV. The IC50 values of chlorella extract, chlorella extract nanoparticles, and peel off gel mask preparations were 83,149 ppm (strong antioxidant), 15,455 ppm (very strong antioxidant), and 155,74 ppm (moderate antioxidant). Masalah penuaan kulit sudah banyak terjadi dikalangan masyarakat yang disebabkan oleh radikal bebas. Oleh karena itu dibutuhkan suatu sediaan kosmetik yang dapat mengatasi penuaan kulit. Mikroalga klorella (Chlorella vulgaris) memiliki aktivitas antioksidan alami yang dapat berasal dari metabolit sekunder seperti flavonoid dan fenolik yang dapat digunakan sebagai anti penuaan. Penelitian ini diawali dari proses ekstraksi serbuk klorella menggunakan metode Ultrasound Assisted Extraction (UAE) dengan pelarut etanol 96% dilanjutkan dengan karakterisasi ekstrak, analisis kandungan fitokimia, kromatografi lapis tipis (KLT), dan kadar flavonoid total ekstrak. Selanjutnya ekstrak dibuat dalam bentuk nanopartikel dengan polimer kitosan dan natrium tripolifosfat (NaTPP) dengan konsentrasi 0,1% dan dilanjutkan dengan karakterisasi nanopartikel. Nanopartikel ekstrak klorella digunakan sebagai zat aktif dalam pembuatan sediaan masker gel peel off kemudian dilakukan evaluasi terhadap sediaan masker dan dilakukan pengujian aktivitas antioksidan terhadap ekstrak, nanopartikel ekstrak, dan sediaan masker dilakukan untuk melihat kemampuan aktivitas antioksidan melalui penangkapan radikal bebas secara in vitro menggunakan DPPH (1,1-difenil-2-pikrilhidrazil). Ekstrak klorella memiliki persen rendemen sebesar 5,18% dan mengandung senyawa alkaloid, flavonoid, steroid, saponin, dan fenolik. Hasil pengujian KLT menunjukkan bahwa adanya senyawa metabolit sekunder yang berpotensi sebagai antioksidan. Kadar total flavonoid ekstrak yang terkandung dalam ekstrak sebesar 67,485 mg/g. Hasil karakterisasi nanopartikel ekstrak menunjukkan persen tramsittan sebesar 87,4%, ukuran partikel 549,5 nm, PDI (Poly Dispersity Index) 0,375 dan zeta potensial +43,35 mV. Nilai IC50 dari ekstrak klorella, nanopartikel ekstrak klorella, dan sediaan masker gel peel off yaitu masing-masing 83,149 ppm (antioksidan kuat),15,455 ppm (antioksidan sangat kuat), 155,74 ppm (antioksidan sedang).
