Abstract

Cempedak bark (Artocarpus integer (Thumb.) Merr.) contains flavonoid including Artoindonesianin-E and Heteroflavon-A as antioxidants. Which can be obtained using the extraction process. The lacks of conventional extraction are requiring a longer time and a large number of solvents, to overcome these limitations, modified extractions have been developed including microwave-assisted extraction (MAE), and ultrasonic extraction (Ultrasound-assisted extraction/UAE). This study aims to determine the solvent concentration and extraction time in UAE and the best extraction power and time in MAE in producing total flavonoid and antioxidant activity. This research was conducted by varying the solvent concentration (30, 70, 96%) and extraction time (20, 50, 80 minutes) for UAE and varying the power (90, 180, 360 Watt) and extraction time (10, 25, 40) for MAE using DX designer, as well as maceration extraction using 70% ethanol concentration and extraction time of 25 minutes. Determination of the best extraction conditions was carried out using a factorial design. Based on the analysis of Design Expert 12®, the solvent concentration factor had no effect and the extraction time factor affected the total flavonoid content and antioxidant activity of UAE, while MAE had no effect on the total flavonoid content and the antioxidant activity of the cempedak bark. The best UAE extraction conditions were obtained at 70% ethanol concentration and extraction time of 80 minutes with a total flavonoid content value of 301.506 mg/100g and antioxidant IC50 value of 43.572 g/mL, and the best condition of MAE at 180 Watt power and extraction time of 25 minutes with a value of total flavonoid content of 442.448 mg/100g and antioxidant IC50 of 102.089 g/mL which were selected based on the highest desirability value. The results of maceration with 70% ethanol concentration and extraction time of 25 minutes obtained total flavonoid content of 503.322±2.422 mg/100g and IC50 of 42.989±6.479 g/mL.
