Abstract

Gambier leaf is natural ingredient that are rich in antioxidants and have potential as antidiabetic because rich of phytochemical compounds with catechins as the dominant phytochemical compounds. However, gambier leaf extract has low bioavailability and solubility. Therefore, gambier leaf extract is formulated into SNE (self-nanoemulsifying) which can increase the bioavailability and solubility of gambier leaf extract in the body so that its potential can be maximized. Stability is an important parameter in the formulation and development of pharmaceutical preparations. Therefore, this study aims to determine the properties and characteristics of the stability of SNE of gambir leaves extract in storage with a predetermined storage period and conditions to obtain a stable pharmaceutical preparation. The results obtained include gambir leaf extract with a yield of 17.32% through the UAE extraction method (ultrasound-assisted extraction). This research use Design-Expert 12® software through a multilevel categoric to obtain 12 SNE formulas for gambier leaves. All SNE formulas of gambir leaves extract have stable organoleptic properties as indicated by stable organoleptic and no precipitation and phase separation in real-time and freeze-thaw. Formulas 3 and 8 in the centrifugation test showed precipitation so that they became unstable formulas in the centrifugation stability test. All formulas have a stable infrared spectral pattern with a constant functional group and no shift or loss of functional groups in real-time for 30 days and freeze-thaw for 6 cycles. Chemometric analysis showed that almost all SNE formulas of gambir leaf extract in real-time and freeze-thaw stability testing real-time and the same cycle in the freeze-thaw grouped in the same group so that they have the same stability characteristics. Keyword: Self-nanoemulsifying, gambier leaves, stability, FTIR-ATR, chemometric Daun gambir menjadi bahan alam yang kaya akan antioksidan dan berpotensi sebagai antidiabetes dikarenakan kaya akan senyawa fitokimia dengan katekin sebagai senyawa fitokimia dominannya. Namun, ekstrak daun gambir memiliki biovailabilitas dan kelarutan yang rendah. Maka dari itu, ekstrak daun gambir diformulasikan kedalam SNE (self-nanoemulsifying) yang dapat meningkatkan bioavailabilitas dan kelarutan ekstrak daun gambir didalam tubuh sehingga potensinya dapat lebih maksimal. Stabilitas menjadi parameter yang penting pada proses formulasi dan pengembangan sediaan farmasi. Maka dari itu, penelitian ini bertujuan untuk mengetahui sifat dan karakteristik stabilitas SNE ekstrak daun gambir dalam penyimpanan dengan jangka waktu penyimpanan dan keadaan yang telah ditentukan untuk mendapatkan sediaan farmasi yang stabil. Hasil yang diperoleh diantaranya ekstrak daun gambir dengan rendemen sebesar 17,32% melalui metode ekstraksi UAE (ultrasound-assisted extraction). Rancangan formula dengan piranti lunak Design-Expert 12® melalui tahap multilevel categoric sehingga diperoleh 12 formula SNE daun gambir. Seluruh formula SNE ekstrak daun gambir memiliki organoleptik yang stabil yang ditunjukkan dengan organoleptik yang stabil dan tidak menunjukkan adanya pengendapan serta pemisahan fase pada pengujian real-time dan freeze-thaw. Formula 3 dan 8 pada uji sentrifugasi menunjukkan adanya pengendapan sehingga menjadi formula yang tidak stabil pada pengujian stabilitas sentrifugasi. Seluruh formula memiliki pola spektra inframerah yang stabil dengan memiliki gugus fungsi yang konstan dan tidak mengalami pergeseran maupun hilangnya gugus fungsi pada pengujian stabilitas real-time selama 30 hari dan freeze-thaw selama 6 siklus. Analisis kemometrik menunjukkan bahwa hampir seluruh formula SNE ekstrak daun gambir pada pengujian real-time dan freeze-thaw mengelompok dalam 4 kelompok dengan hasil bahwa tiap sampel pada tiap minggu penyimpanan pada pengujian stabilitas real-time dan siklus yang sama pada pada pengujian freeze-thaw mengelompok pada kelompok yang sama sehingga memiliki karakteristik stabilitas yang sama.
