Abstract

Termites are pests that can cause damage to various types of wood, buildings and plants. Termites can digest cellulose because their intestines contain cellulolytic bacteria. Cellulolytic bacteria from termite digestion have been isolated, two of which are Enterobacter Clocoae and Klebsiella Pneumoniae. This study aimed to study the effect of pH and temperature on cellulase enzyme activity isolated from Enterobacter Clocoae bacteria and Klebsiella Pneumoniae bacteria obtained from the digestive tract of termites. Crude extract of cellulase enzyme was obtained extracellularly from Enterobacter Clocoae and Klebsiella Pneumoniae bacterial cultures. Extracellular enzymes were obtained from the process of centrifugation of bacterial liquid cultures at a speed of 15,000 rpm at 4 oC for 5 minutes. The crude extract enzyme was then fractionated with ammonium sulfate. The fraction with the highest enzyme activity was used to determine the effect of pH and temperature on cellulase enzyme activity. Enzyme activity was determined using CMC substrate and the result of hydrolysis in the form of sugar was determined by the 3,5- dinitrosalicylic acid (DNS) method and measured by UV-Vis spectrophotometer. The results showed that the extracellular enzyme activity of the crude extract obtained from Enterobacter cloacae was 0.001 U/mL, while the extracellular enzyme activity obtained from Klebsiella pneumoniae was 0.0007 U/mL. The highest enzyme activity of Enterobacter cloacae was obtained in fraction 2 with cellulase enzyme activity of 0.017 U/mL, while the activity of cellulase enzymes produced from Klebsiella pneumoniae which had the highest activity was found in fraction 3 with enzyme activity of 0.012 U/mL. Cellulase enzyme activity of Enterobacter cloacae optimum at pH 5 resulted in enzyme activity of 0.020 U/mL and optimum at 35oC with cellulase enzyme activity of 0.018 U/mL. Cellulase enzyme activity of Klebsiella pneumoniae optimum at pH 8 resulted in enzyme activity of 0.016 U/mL and optimum at 30oC with cellulase enzyme activity of 0.018 U/mL. Based on the data obtained, it can be concluded that Enterobacter cloacae has the best conditions at pH 5 and a temperature of35oC, while Klebsiella pneumoniae has the best conditions at pH 8 and a temperature of 30oC.
