Abstract

Carica papaya Linn. ethanol leaves extract contains saponin, flavonoid and polyphenol as antibacterial, antiinflamation, and antioxidant used for burn therapy. Formulation of spray gel aims to reduce contamination or infection and pain when applied to burn patients. Spray gel was formulated by carbopol®940 and gellan gum as factors used 22 factorial design Design Expert®10 program. The influence of factor composition and its interaction observed by the analysis result of organoleptis, homogeneity, viscosity, pH, adhesion, washability, spraying pattern, pump delivery, and hedonic. Result were analyzed by DX®10 to determine the optimum formula. Optimum concentration of 1.2% carbopol®940 and 0.218% gellan gum had diameter of pattern of spraying 1.644 ± 0.0196 cm, pH 5.58 ± 0.0058, adhesion on skin 2.870 ± 0,04 second, sticking paper filter 2.813 ± 0.0651 sec, washability 3.067 ± 0.0577 mL, pump delivery 34.742 ± 0.0006, and hedonic 3.515 ± 0.0212. Carbopol®940 increased adhesion and washability, but decreased pH, spraying pattern, pump delivery, and hedonic. Gellan gum increased spray pattern, pump delivery, adhesion, washability, and hedonic but decreases pH response. Saponin content of Carica papaya Linn. ethanol leaves extract of 13.851 ± 0.208%. Measurement of %diffused optimum formula and pure solution of extract on the 360th minutes (6 hours) are 18.354 ± 0.103% and 25.867 ± 0.113%. Compartmental analysis was observed using WinSAAM® resulting optimum formula follows lag compartment model with correlation between Qo and Qc proven by p-value of <0.05. Study of stability using heating/cooling method at 4 and 45oC every 24 hours during 3 days show that optimum formula spray gel is physically stable, but there is a decrease in saponin content. Interaction study between papaya leaf extract and bases using FTIR shows nothing chemical interaction was occurred. The conclusion of the result is Carica papaya L. ethanol leaves extract with bases carbopol®940 and gellan gum can be formulated into spray gel.
