Abstract

Noni fruit (Morinda citrifolia L.) contains several secondary metabolites, one of which is flavonoids. Flavonoids themselves have a special compound, namely quercetin which has the potential as an immunomodulator. Traditional medicinal plants have low solubility in fat and permeability are less able to penetrate the absorption barrier, thus affecting the bioavailability of these natural compounds in the body. Orally dissolving film technology can be used as an alternative to increase permeability. Orally dissolving film of noni fruit extract will be formulated based on changes in the concentration of pullulan polymer and propylene glycol using the Design-Expert® with the factorial design method which produces 4 formulas with different concentrations and determines the thickness response, folding resistance, disintegration time and percent elongation to obtain orally dissolving film of noni fruit extract with physical conditions that meet the requirements, in order to obtain the best formula. The best formulation obtained was evaluated for pH, weight uniformity, content uniformity, and stability tests. In addition to flavonoids, phytochemical screening of noni fruit extract produced alkaloids, flavonoids, and saponins. The total flavonoid content of the noni fruit extract produced was 410,93 mgCe/g extract. The optimum formulation of noni fruit extract used pullulan and propylene glycol at concentrations of 364.866 mg and 60 mg, respectively. The optimum formulation has a thickness of 0.128 mm, a folding resistance of 466, a disintegration time of 33 seconds, and an elongation percent of 34%. The optimum formula pH was 4.75, the weight uniformity was 0.081 mg, the concentration uniformity was 107.78 ± 3.829%, and the organoleptic remained good after the stability test was carried out by the cycling test and good stability in the % recovery indicated by the sig value > 0.05 using themethod Paired T-Test.
