Abstract

Wounds are a form of tissue damage to the skin and wound healing can be accelerated by treating the wound. Rosella (Hibiscus sabdariffa L.) plant can be used as a wound healer which contains flavonoid compounds, tannins, and saponins. This study aims to determine the effect of variations in the concentration of ethanolic extract of rosella petals on the characterization of spray gel preparations and open wound healing in Sprague Dawley male white rats. The treatment groups used in this study were 8 treatments, including; negative, positive, placebo, pure extract groups and variations in the concentration of spray gel ethanol extract of rosella flower petals by 10% (F1), 15% (F2), 20% (F3) and 25% (F4). Extract characterization included organoleptic test, water soluble extract content, ethanol soluble extract content, metal contamination, microbial contamination, water content, total ash content and acid insoluble total ash content. Evaluation of spray gel preparations included organoleptic tests, homogeneity, pH, viscosity, adhesion, spraying patterns, pump delivery, washability and stability. Open wounds were made using a biopsy punch with a diameter of ±8 mm and a depth of ±2 mm. Wound observations were carried out on days 4, 8, 12, 16 and 20 for 20 days and the wound diameter was measured and analyzed using imageJ and SPSS applications. The results of the characterization of the ethanolic extract of rosella flower petals obtained that all tests met the standards of the Indonesian Herbal Pharmacopoeia (2017). The results of the evaluation of the spray gel of rosella flower petals ethanol extract obtained that all tests met the requirements. The fastest wound healing is influenced by variations in the concentration of ethanol extract of rosella petals where the more concentration used, the faster the wound healing process. Based on the results of the study, it can be concluded that the best formula as a wound healer is in formula 4 with a concentration of 25% and healing time for 12 days.
