Abstract

Background: Traumatic ulcer are lesions of the oral mucosa caused by thermal, mechanical and chemical. Long-term use of topical medication can lead resistance to fungal infection. Several studies have shown that patin fish skin contains glycine and glutamic acid which can increase macrophage activation and fibroblast proliferation in the wound healing process that wound healing will be accelerated. Aim : to determined the effect of gelatin skin catfish to increasing macrophage and fibroblast and decreasing the diameter of traumatic ulcer. Method: A true experimental laboratories with postest only control group design. 24 wistar strain male rats were devided into 4 groups. Ulcer was made on the labial mucosa of mandibular by thermal induction. Ulcer was tread with placebo gel and gelatin skin catfish with 10%, 20% and 30% concentration for 7 days. Rats were euthanazied on days 3,5,7 and ulcerated mucosal tissue was biopsied to count the number of macrophage and fibroblast using olympus software. Data were analyzed using IBM SPSS statistic 25 version. Result: Gelatin skin catfish with 10%, 20% and 30% concentration had a difference in mean from the 3rd, 5th and 7th days. Based on Npar test there were significant differences in number of macrophages cells between the gelatin skin catfish treatment groups with 10%, 20% and 30% concentration. Based on post hoc test there was significant difference in the number of fibroblast cell gelatin skin catfish with 10%, 20% and 30% concentration compared to placebo gel (p<0,05). Based on ulcer diameter measurement gelatin skin catfish with 10%, 20% and 30% concentration was able to decreasing the diameter of ulcer. Conclusion : Gelatin skin catfish with 10%, 20%, 30% concentration there was a significant difference in the number of macrophage cells and increased the number of fibroblast cell from the 3rd, 5th, 7th days and decreased the diameter of ulcer.
