Abstract

Research has been carried out to study the properties of tea leaf catechins prepared from chitosan-sodium alginate polymer by ionic gelation method using a nanoparticle delivery system. This study aims to determine the effect of variations in the concentration of sodium alginate of each formula of 1.6 mg; 3.2 mg; and 4.8 mg on the resulting nanoparticles. The optimum formula will be determined based on the highest %EE value, and also carried out stability test using the heating-cooling cycle method and particle characterization. Tea leaf extract contains secondary metabolites including flavonoids, phenolics, tannins, saponins, alkaloids, and triterpenoids. The total flavonoid content of tea leaf extract was 380,333 mg CE/g extract. Manufacture of nanoparticles using the ionic gelation method by dispersing tea leaf extract in a chitosan solution, then drop by drop sodium alginate and CaCl2 as a crosslinker. The optimal formula (F3) of nanoparticles has %EE 96.852% with characterization of particle size 328.3 nm; PDI value 0.3819; and zeta potential +26.79 mV. TLC analysis showed that the extract and nanoparticles of tea leaves containing catechin compounds with an Rf value of 0.74. The results of the stability test of nanoparticles of the tea leaf extract nanoparticles showed organoleptic changes with the formation of precipitates and a change in color to cloudy orange, having a pH of 5.413; 5,470; 5,247 with a percentage reduction of 5,657%; 4.776%; 3.622% for each formula. It can be concluded that the tea leaf extract nanoparticles have good characterization and stability.
