Abstract

Diarrheal disease caused by Escherichia coli bacteria is still common in Indonesia. Kersen plant (Muntingia calabura L.) is widely used to treat various diseases, namely jaundice, gout and cough, but it can also be used as an antipyretic, antioxidant, anti-inflammatory, antidiabetic, antibacterial and antiulcer. Kersen leaves contain active compounds of flavonoids and tannins which act as antibacterial and anti-diarrheal. This study aims to evaluate the ethyl acetate fraction of cherry leaves as an antidiarrheal. This research was conducted using Wistar strain male white rats induced by Escherichia coli bacteria. Mice were divided into 7 groups, namely the normal group, negative control (1% NaCMC), positive control (Gentamycin), and four test groups with doses of 9.4, 18.8, 37.6 and 75.2 mg/kg BW. The initial time parameters for diarrhea occurred from 24 to 30 hours. Parameters of stool weight, stool consistency, frequency of diarrhea, body weight, number of Escherichia coli bacteria colonies in rat feces in the test group at a dose of 75.2 mg/kg has an effect close to the control group positive. The results also showed that the antidiarrheal effect of a dose of 9.4 mg/kgBW was 36.20%, a dose of 18.8 mg/kgBW was 41.00%, a dose of 37.6 mg/kgBW was 43.44% and a dose of 75.2 by 50.19%. This shows that the antidiarrheal effect of the ethyl acetate fraction of cherry leaves at a dose of 75.2 mg/kg BW was not significantly different (p>0.05) to the positive control (gentamicin). The total flavonoid test results were 56.99 mgQE/g or 5.699% and the total tannins were 165.29 mgTEA/g or 16.529%. The effective dose (ED50) of the ethyl acetate fraction of cherry leaves as an antidiarrheal is 72.48 mg/kgBW.
