Abstract

Benzoic acid has antimicrobial activity, but poorly soluble in water. Various strategies are currently being developed to get better drug properties in terms of solubility and stability. Co-crystal is a combination of two or more crystals that are intended to improve the intrinsic properties of an water-soluble drug substance. The advantages of co-crystal products compared to other forms are seen from their superior physicochemical properties, solubility, bioavailability, and thermal stability. The development of the use of PVA now has been used in drug formulations. PVA is a water-soluble synthetic polymer which is used as a surfactant and stability agent. PVA has a better ability to dissolve water insoluble drugs and generally non-toxic to biological membranes. The purpose of this study was to determine the effect of benzoic acid with PVA conventional coating and bead milling on physical and chemical benzoic acid/PVA Co-crystal characteristics. The co-crystal manufacturing method chosen is grinding with Bead Milling because it is more economical and environmentally friendly. Evaluations were carried out on Benzoic Acid/PVA co-crystal samples included physicochemical properties tests including pH tests, viscosity tests, solubility tests, thermodynamic stability tests, mechanical stability tests, and X-ray diffraction structural characteristics tests and FTIR tests. The results of the evaluation of Benzoic Acid/PVA co-crystals prepared by Bead Milling showed that there were effects such as increased solubility, significant changes in pH, decreased viscosity, good thermodynamic and mechanical stability, also physical interactions in the FTIR spectra and decreased diffractogram intensity. Comparison of the stability of Benzoic Acid/PVA co-crystals prepared by Bead Milling showed better results than the stability of pure drug components and mixtures thereof.
