Abstract

Background: Xanthine oxidase is an enzyme that catalyzes the formation of uric acid. Secondary metabolites that have a potential as xanthine oxidase inhibitor or antihyperuricemia are flavonoids, alkaloids, terpenoids, saponins, and tannins. From the latest research, ethanol extract of the stem of kayu kuning contains secondary metabolites such as flavonoids, alkaloids, terpenoids, saponins, and tannins, so it has antihyperuricemic potential. Secondary metabolites, such as flavonoids and alkaloids are generally polar. In principle, a compound dissolves in solvents that have the same level of polarity, so ethanol is used because it has the characteristics of a polar solvent. This research was conducted to determine the ability of the ethanol fraction of yellow wood stems (Arcangelisia flava Merr) as an inhibitor of the xanthine oxidase enzyme. Method: The type of the research is an in vitro experiment. In this research, phytochemical tests and inhibition tests were carried out. The test of ability inhibition using 4 concentration such as 6,25 ppm, 12,5 ppm, 25 ppm, and 50 ppm of ethanol fraction of the stem of kayu kuning and allopurinol as a comparison, then measure the absorbance using UV-Vis spectrophotometry with 293 nm of wavelength. Results: Ethanol fraction of the stem of kayu kuning contains secondary metabolites such as alkaloid, flavonoids, terpenoids, and quinones, but not contain steroids, saponnins, and tannins. Ethanol fraction of the stem of kayu kuning has IC50 value as 31,2729 ppm and classified as strong ability to inhibition xanthine oxidase enzyme.
