Abstract

Background: Insomnia is a sleep disorder that decreases quality of life and melatonin level due to chronic inflammatory, but the current treatment is still not effective. Cinnamon is rich in anti-inflammatory and antioxidants compounds. This study aims to determine the effect of the cinnamon nonpolar fraction on melatonin levels. Methods: This study is an experimental study in vivo post-test only control group at Eureka Research Laboratory. Thirty male Wistar rats were divided into six treatment groups: normal control, negative control (Na-CMC 0.5%), positive control (estazolam 0,1 mg/kgBW), and cinnamon nonpolar fraction group (CNF) 5 mg/kgBW, 10 mg/kgBW, and 20 mg/kgBW. CNF was obtained through separation funnel with n-hexane solvent from macerated cinnamon simplicia with 96% ethanol. Except normal control, rats were induced insomnia with intraperitoneal administration of 300 mg/kgBW p-chlorophenylaline (PCPA) for two days. The administration of experiment materials were performed with intragastric sonde for 14 days and after that the rats were euthanized and their hypothalamic tissue was taken to measure the melatonin level with enzyme-linked immunosorbent assay (ELISA). The data were then analyzed with SPSS by univariate, bivariate, and multivariate manner with a statistical significance of p < 0.05. Results: Every CNF group has higher melatonin levels than negative control. The increase of melatonin level in the groups that got CNF was all statistically meaningful and the CNF dose of 20 mg/kgBB provided melatonin levels that exceeded normal control levels. Conclusion: CNF had an effect in increasing hypothalamic melatonin levels in PCPA-induced insomnia rats.
