Abstract

Bacteria Escherichia coli is a gram-negative bacteria, whereas Staphylococcus aureus bacteria is a gram-positive bacteria. Both of these bacteria are used to represent the microorganisms in testing the activity of antibacterial compounds that provide a different sensitivity when spreading in the body. The aim of this research is to know the antibacterial activity of fractions and active compounds from leaves extract (Syzygium antisepticum Blume.) to Escherichia coli and Staphylococcus aureus. The research has finished carried out from October 2017 to February 2018. The method used in this research is maceration extraction, liquid-liquid fractionation, antibacterial activity test and determination of minimum inhibitory concentration by agar diffusion method and isolation of active compound by column chromatography method. Tested bacteria used were Escherichia coli and Staphylococcus aureus. Data are presented in table based on the average of inhibitory diameter and standard deviation. The results of this study indicate that the fraction of methanol water is active against the tested bacteria. From the fraction of methanol water obtained an antibacterial compound, from the bottle 5 which is suspected of tannin compound with a value of Rf 0,6. The minimum inhibitory concentration from the fraction of methanol water of ubah leaf extract was 500 μg/ml would against Escherichia coli and 250 μg/ml against Staphylococcus aureus. The minimum inhibitory concentration of the active compound was 125 μg/ml would against Escherichia coli and 62.5 μg/ml against Staphylococcus aureus. Fraction and acive coumpounds of ubah leaves extract have antibacterial activity against Escherichia coli and Staphylococcus aureus with the active compounds is tannin.
