Abstract

Klindamisin bekerja dengan meghambat sintesis protein bakteri yang dimodifikasi dalam bentuk transfersom memiliki kemampuan deformabilitas yang dapat meningkatkan elastisitas melalui destabilisasi lipid bilayer. Karakterisasi transfersom klindamisin dengan variasi edge activator surfaktan Tween 80 dan Span 80 melalui program Design-Expert 12® metode Simplex Lattice Design dengan level 0-10 % untuk melihat pengaruh masing-masing faktor ataupun interaksi antar kedua faktor terhadap karakterisasi transfersom yang dihasilkan. Parameter evaluasi meliputi % efisiensi penjerapan, ukuran partikel, indeks polidispersitas, dan nilai zeta potensial, Formula optimum dilanjutkan uji stabilitas menggunakan metode stabilitas ICH Q1(R2) dan asean guideline for stability study. Hasil karakterisasi menunjukkan kombinasi tween 80 dan span 80 dapat menurunkan nilai %EE, memperkecil ukuran partikel, PDI, dan zeta potensial. Konsentrasi surfaktan yang digunakan untuk menghasilkan formula optimum menggunakan konsentrasi span 80 sebanyak 10% dan tween 80 sebanyak 0%. Hasil uji stabilitas transfersom formula optimum menunjukkan masa simpan pada suhu 5oC hasil ekstrapolasi dari ketiga kondisi penyimpanan berbeda yaitu selama 175,79 hari.
