Abstract

Oil palm waste is the residue from the oil palm plant which is not included in the main product or can also be referred to as a by-product of palm oil processing. Palm oil liquid waste that is wasted into the aquatic environment can later cause pollution. Pollution that occurs is a result of the formation of a layer of fat on the surface of the water so that it can inhibit the solubility of oxygen contained in the water and can disturb the organisms that are around. One way that can be done in the processing of palm oil liquid waste is by using the bioremediation method using the help of biological agents in the form of bacteria to degrade lipids in palm oil liquid waste. Bacteria that have the ability to degrade lipids are classified into lipolytic bacteria. Lipolytic bacteria have the ability to produce lipase enzymes which can catalyze the hydrolysis of fats into fatty acids and glycerol. The lipase enzyme produced by bacteria by lipolytic is able to decompose organic matter in the form of fat quickly by breaking the ester bond of triglycerol into fatty acids and glycerol which are soluble in air. This study aims to determine the genus of bacteria that have high lipase activity isolated from palm oil wastewater. This research was conducted from December 2022 to March 2023, taking place at the Microbiology Laboratory and Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University, Indralaya. The stages of the research were isolation, purification, qualitative lipase activity test, inoculum preparation, lipase enzyme production, lipase activity test quantitatively, characterization and assistance. The conclusion of the study was that the isolates of lipolytic bacteria from palm oil wastewater which had the ability to produce lipase enzymes amounted to 15 bacterial isolates from 27 bacterial isolates. The bacterial isolates LCK5A, LCK5E, and LCK2G had high lipase activity values of 2.1175 U/ml, 1.7375 U/ml, and 1.625 U/ml based on quantitative tests, respectively. Bacterial isolates LCK5A, LCK2G were identified as the genus Citrobacter and isolates LCK5E were identified as the genus Lactobacillus.
