Abstract

Catechins compounds in gambier leaves have an antioxidant effect. The purpose of this study is to determine the optimal results of gambir leaf extract and the characteristic of optimal gambier leaf extract. The extraction method used in the study was ultrasoundassisted extraction (UAE). Data analysis was carried out using software design expert 13 with factorial 23. Factors in the extraction process used are times (10-30 minutes), temperature (20-500C), and material: solvent ratio (1:7-1:10). The response of the observed study results consisted of % extract yield, total flavonoid contents (TFC), and antioxidant activity (IC50) of the extract. The optimal process factor condition results are selected based on the condition with the highest desirability value. The optimal results of gambier leaf extract based on the extraction process factor were obtained in the extraction treatment time 30 minutes, temperature 500C, with a ratio of material: solvent 1:10 which % yield extract 7,8%; total flavonoid contents 467,514 mgCE/g; and antioxidant activity value of 66,013 ppm with a desirability value 0,998. The characteristics of the optimal gambier leaf extract have a moisture content 0,13% and a drying shrinkage 0,15%. The phytochemical screening results of optimum gambier leaf extract wer found positif for phenolic, flavonoids, and tannins. The results of verification optimal condition of extract obtained are in the range of 95% PI low and 95%PI high which indicates that there is a conformity to the predicted value with the value of observations that have been made.
