Abstract

Jackfruit (Artocarpus heterophyllus) is a medicinal plant that is rich in phenolic and flavonoid compounds. Phenolic compounds are known to have high potential as antioxidants. Compounds that act as antioxidants generally also have the potential to act as anti-inflammatory. This study aims to determine the in vitro antioxidant activity and in vivo anti-inflammatory activity of A. heterophyllus leaf fractions. The research began with extraction using the maceration method using solvents with varying polarities (n-hexane, ethyl acetate and methanol). Each fraction was tested for flavonoid and total phenolic content using the colorometric method and antioxidant activity using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. The fraction that showed the highest antioxidant activity was then tested for anti-inflammatory activity in vivo by measuring the volume of edema formed from carrageenan induction. Testing was carried out with 3 dose variations of 200, 400, and 800 mg/KgBW. The research results showed that the methanol fraction had the highest total phenolic content and antioxidant activity compared to other fractions with a total phenolic value of 50.31 mgGAE/g and antioxidant activity with an IC50 value of <15.6 ppm. Testing of the anti-inflammatory activity of the methanol fraction showed the highest anti-inflammatory activity at a dose of 800 mg/KgBW with a percent inflammation inhibition value of 54.34%, while the positive control aspirin was 40.42%. Statistical analysis showed that there was no significant difference between the test group treated with doses of 200, 400, and 800 mg/KgBW and the positive control group. Based on the data, it was concluded that substances that are active antioxidants are also active as anti-inflammatories and the methanol fraction of jackfruit leaves has the potential to be developed as a raw material for anti-inflammatory drugs.
