Abstract

Cikori root flour (Cichorium intybus L.) is a functional food containing inulin and is useful as a prebiotic in stimulating the growth of Bifidobacterium longum (B. longum). Prebiotic cikori root flour is made into an orally dissolving film (ODF) with the advantage of fast onset without interfering with the release of active ingredients with a pleasant taste. Research was conducted to see the prebiotic potential of cikori root flour in ODF to increase the growth of B. longum and to test the antibacterial metabolites against Escherichia coli (E. coli). Cikori root flour was tested for quality according to SNI 01-3751-2009. ODF was made using the solvent casting method with 3 formulas with variations of cikori root flour, namely 1%, 2% and 5% w/w. The best formula was tested for B. longum growth using the total plate count (TPC) method. The metabolites were tested as antibacterial against E. coli using paper disk diffusion. The results of the quality standardization analysis of cikori root flour meet the requirements of SNI 01-3751-2009. The observation results showed that the third formula was the best formula with 5% w/w of cikori root flour. B. longum which grew after being treated with ODF F3 was 1.110×1010 CFU/mL. The effect value and prebiotic index of ODF F3 are 10.034 and 0.967, indicating its potential as a prebiotic. Antibacterial testing against E. coli from B. longum metabolites stimulated by ODF F3 showed an inhibitory power of 7.4±0.624 mm with medium criteria. Conclusion ODF F3 is the best formula with prebiotic potential to stimulate B. longum so that it produces metabolites to inhibit E. coli with moderate effectiveness criteria.
