Abstract
Sambiloto leaf extract (Andrographis paniculata) contains secondary metabolites, one of which is flavonoid. Flavonoid have a special compound, quercetin, which has the potential to stimulate probiotic and antibacterial activity. The extract has a bitter taste so that it is formulated in the form of orally dissolving film (ODF) preparation which is expected to cover the bitter taste and provide an attractive appearance. This study aims to determine the effect of ODF preparation of sambiloto leaf extract to stimulate probiotic Bifidobacterium longum (B. longum) in inhibiting Escherichia coli (E. coli). Extraction of sambiloto leaves using 96% ethanol solvent by maceration method. ODF formulation with solvent casting method. Probiotic growth test of B. longum with Total Plate Count (TPC) method and antibacterial activity test with paper disc diffusion method. The results of the sambiloto leaf extract obtained had a yield of 13,508%. The results of preparation evaluation obtained formula 2 as the best formula (p value <0,05) with weight uniformity %CV of 2,671%; film thickness 0,102 ± 0,008 mm; preparation pH 6,240 ± 0,026; percent elongation 22,250 ± 1,372%; folding endurance 443 ± 2; disintegration time 16,633 ± 0,822 seconds; and uniformity %recovery 100%. The best B. longum probiotic growth test results (p value <0,05) were found in the best ODF-stimulated probiotic with a %increase of 22,726%. The antibacterial activity of the combination of probiotic metabolites and the best ODF (p value <0,05) showed a moderate category with an inhibition zone of 7,5 mm. Based on the results of the study, it can be concluded that formula 2 is the best formula that has the potential to stimulate B. longum so that it can inhibit E. coli.
