Abstract

Fraksi etil asetat daun singkong mengandung senyawa rutin yang efektif menghambat pertumbuhan bakteri Staphylococcus aureus. Penelitian ini bertujuan mengetahui variasi konsentrasi kalsium klorida sebagai crosslinker terhadap transdermal patch yang dihasilkan dan mengetahui aktivitas antibakteri sediaan transdermal patch. Polimer yang digunakan pada penelitian ini adalah kitosan dan natrium alginat. Pembuatan transdermal patch dibuat dengan metode pencampuran (solvent casting) dalam 3 formula yakni FI, FII dan FIII dengan variasi konsentrasi kalsium klorida 0,5%, 0,75% dan 1%. Hasil formula terbaik didapatkan konsentrasi kalsium klorida 0,5% pada FI yang menghasilkan patch dengan permukaan halus, elastis dan transparan. Formula terbaik (FI) sediaan transdermal patch memiliki persen elongasi 80,116%, pH 6,72, keseragaman bobot 48,533 mg, kelembapan <4%, uji swelling 8.7471, ketebalan 0,117 mm, keseragaman kadar 100.008% ± 0.221. Hasil uji stabilitas menunjukkan bahwa sediaan transdermal patch baik dan stabil selama 6 siklus (p>0,05). Transdermal patch fraksi etil asetat daun singkong dengan konsentrasi kalsium klorida 0,5% memiliki karakterisasi dan stabilitas yang cukup baik. Transdermal patch fraksi etil asetat daun singkong menghasilkan diameter zona hambat sebesar15,86 ± 1,026 mm dengan kategori daya hambat kuat dan memiliki perbedaan signifikan terhadap kontrol positif (p<0,05).
