Abstract

Temu mango rhizome (Curcuma amada Roxb.) is one of the plants that has anti-inflammatory and antidiabetic activities. The safety of using temu mango rhizome as a natural medicinal preparation must be supported by scientific research on toxicity testing. This study aims to evaluate the characteristics of acute toxicity of ethanol extract of temu mango rhizome to female Wistar rats using the Acute Toxic Class method. The test results showed that the extract met the standards set, namely organoleptical solid brown color, viscous form, distinctive aroma of the extract, moisture content 7 ± 1, water soluble juice content 53.33 ± 5.77, ethanol soluble juice content 83.33 ± 5.77, total ash content 2.16 ± 0.28, acid insoluble ash content 0.68 ± 0.28, and specific gravity 0.99 ± 0.02. Toxicity test observations showed that no deaths occurred but toxic symptoms such as weakness were seen in the test animals. The administration of ethanol extract of temu mango rhizome at a dose of 10,000 mg/kgBB showed the effect of macroscopic and microscopic organ damage. Based on the average data before and after administration of the extract there was a significant decrease in the weight of the test animals (p<0.05). In the examination of biochemical parameters, there were no significant differences in the levels of SGOT, SGPT, creatinine, ureum, LDL, and triglycerides (p>0.05). The average levels of biochemical parameters of the treatment group were SGOT 127.96 ± 17.70 U/L, SGPT 47.98 ± 2.62 U/L, creatinine 0.46 ± 0.03 mg/dL, ureum 21.32 ± 1.91 mg/dL, LDL 6.57 ± 2.78 mg/dL, and triglycerides 100.09 ± 47.04 mg/dL. The LD50 value of temu mango rhizome extract is 10,000 mg/kgBB. Based on the results of the study, it is concluded that the acute toxicity test of ethanol extract of temu mango rhizome against female Wistar rats is practically non-toxic.
