Abstract
Pacar kuku (Lawsonia inermis) is a traditional medicinal plant that is rich in phenolic compounds which are generally active as antioxidants. Measuring antioxidant activity can be done using various methods. In this study, the antioxidant activity of L. inermis leaves were determined using the DPPH (2,2-diphenyl-1-picrylhydrazyl), FRAP (Ferric Reducing Antioxidant Power) and FTC (Ferric Thiocyanate) methods. The research began with determining the total of phenolic and flavonoid compounds contained in the fractions (methanol, ethyl acetate, and n-hexane) of L. inermis leaves and determining the antioxidant activity of each fraction. The results showed that the highest levels of phenolic compounds were in the ethyl acetate fraction 49.97 mg GAE/g, while the highest levels of flavonoids were in the methanol fraction 39.22 mg QE/g. The determination antioxidant activity using the DPPH and FRAP methods showed the highest activity in the methanol fraction with an IC50 value of 16.051 mg/L for the DPPH method, and Fe2+ reduction power of 361.52 mg/L with the FRAP method. The antioxidant activity of the methanol fraction was proportional to the total flavonoid content. For the FTC method, the highest was obtained in the ethyl acetate fraction with a % inhibition value of 61.06%. This is directly proportional to the highest total phenolic content in the ethyl acetate fraction.
