Abstract
Sambiloto leaves (Andrographis paniculata (Burm. F) Wall. Ex Ness) contain secondary metabolites with antibacterial, anti-inflammatory, and antioxidant activities, making them a potential agent for burn wound healing. This study evaluated a 1% submicron emulsion of sambiloto leaf extract prepared with a combination of Tween 80 and ethanol at a 25% concentration, testing its burn wound healing activity in Wistar rats. The study included five groups normal, positive control (tekasol), negative control, 1% sambiloto cream, and 1% submicron emulsion. Observations focused on scab formation and wound recovery. The evaluation of the 1% submicron emulsion showed a clear dark green appearance with the characteristic aroma of sambiloto and basil oil, a pH of 6.233±0.057, viscosity of 13.667±0,122cP, specific gravity of 1.034±0.001g/cm3, andrographolide content of 19.636±1.545%, entrapment efficiency of 95.840±0.057%, droplet size of 118.5±8.697nm, polydispersity index of 0.208±0.009, and zeta potential of -24.866±2.635mV with no irritation detected. Scab formation occurred by day 4, except in the negative control (day 10). Based on One Way ANOVA analysis, the submicron emulsion achieved the highest healing value of 94.55±1.47% which was not significantly different from the positive control (90.60±3.28%) but significantly different from the 1% sambiloto cream group (85.05±1.99%). The histopathological analysis of skin tissue treated with submicron emulsions showed increased fibroblasts density, collagen, and normal inflammation, similar to the positive control. Based on this study, the 1% submicron emulsion was more effective in promoting burn wound healing. Keywords: scab, burn wounds, sambiloto, submicro emulsion
