Abstract

Usnic acid is a compound with antibacterial, anti-inflammatory properties, and potential in accelerating burn wound healing. Usnic acid has good bioavailability and low solubility in water so it needs to be formulated in a nanoemulsion system with a combination of citronella oil as the oil phase and cremophor RH-40 as a surfactant. This study aims to determine the effect of variations in the concentration of Cremophor RH-40 and citronella oil on the usnic acid nanoemulsion formula using the Regular Two-Level Factorial Design method and burn wound healing activity test. The optimum formula was determined based on the characterization of entrapment efficiency, viscosity, pH, and specific gravity. The results showed that the optimum formula had a particle size of 236.133 ± 13.4024 nm, a polydispersity index of 0.351 ± 0.01557, and a zeta potential of -15.533 ± 2.2723 mV. The effectiveness of the preparation was tested as a burn wound healing agent in wistar rats with 0.33% usnic acid content. Based on One-Way ANOVA analysis, this nanoemulsion showed no significant difference in effectiveness from the positive control, with % recovery reaching 96.12 ± 1.82% within 12 days. These results indicate that usnic acid nanoemulsion has the potential as an effective pharmaceutical preparation for healing burns. Keywords: Burns, Nanoemulsion, Percent Recovery, Usnic Acid
