Abstract

Avicennia marina leaves have long been used as traditional medicine due to the presence of secondary metabolite compounds that have biological activity, including antioxidant activity. The mangrove forest area in Sungsang IV village is still well maintained and is a place for mangrove restoration and nursery. This study aims to determine the metabolite profile, compound abundance, antioxidant levels and identification of metabolite compound groups in A. marina leaves originating from Sungsang IV, Banyuasin, South Sumatra. The method involved taking mature leaves in sequence 3-5 from the shoots through accidental. Metabolite profile analysis was conducted using a metabolomics approach with GC-MS and antioxidant levels were determined using the DPPH method with quercetin and ascorbic acid as standard antioxidants. The GC-MS analysis data were utilized to trace the compound class and biosynthesis pathway from the total compound abundance to identify compounds with potential bioactivity using the PubChem, ChEBI and PlantCyc websites. Based on the GC-MS chromatogram, the results of the metabolite profile of the 96% methanol extract of A. marina leaves showed a total abundance of 99.07%. A total of 22 compounds were detected, with 4 dominant compounds identified. The dominant compounds were 2-Propanol, 1,1'-oxybis (26.8%) which has antimicrobial bioactivity, 4-Methyl-2,4-bis(p-hydroxyphenyl)pent-1-ene, 2TMS derivative (18.17%) with antioxidant, antibacterial, antimalarial and anticancer activity, and compounds 1-Propanol, 2,2'-oxybis (15.6%) and 1-Propanol, 2-(2-hydroxypropoxy)-. (15.6%) which have bioactivity as antimicrobials. The antioxidant content of A. marina leaves was 39.16 ± 10.05 ppm using ascorbic acid and 64.21 ± 17.32 ppm using quercetin. The group of compounds present in A.marina leaves exhibited bioactivity as antioxidants, antimicrobials, antitumors, anticancer, anti-inflammatory, antifungal, antiasthma, antiandrogenic, antifungal, anticancer, and antimalarial agents, which have the potential to be developed as herbal medicines and fishery preservatives.
