Abstract

Mangosteen peel extract contains alpha-mangostin which accelerates wound healing by stimulating fibroblasts, increasing collagen synthesis, and has anti-inflammatory and antimicrobial properties. Alpha-mangostin transethosomes were developed to improve transdermal delivery because of their better skin penetration and high bioavailability. For ease of use, transethosomes are formulated in gel form. This study aims to obtain the alpha-mangostin transethosomes gel formula with the best gelling agent concentration for wound healing. The formulation of transethosomes gel was carried out by combining carbomer 940 gel base with concentration variations of 0.5%; 1%; 1.5%; 2% and 1% PVA. The results of the evaluation of transethosomes gel based on organoleptic, homogeneity, pH, viscosity, spreadability, adhesiveness, and washability tests showed that the best formula was formula 3 with a concentration of carbomer 940 0.1% and 1% PVA. The average results of the characteristics of pH, viscosity, 150 gram spreadability, 100 gram adhesiveness and washability were respectively 6.12 ± 0.010; 2763.65 ± 44.500 cP; 6.90 ± 0.100 cm; 5.31 ± 0.015 s and 8.26 ± 0.305 mL. The stability test of the best formula using the cycling test method showed that the transethosome gel did not experience organoleptic and syneresis changes, but the gel experienced an increase in pH value and a decrease in viscosity and content values. The results of morphological testing using the transmission electron microscopy method showed that the transethosome vesicles in the gel had a spheric al shape. Diffusion testing showed that there was a difference in the percentage produced from the diffusion test between alpha mangostin extract, alpha mangostin transethosome and alpha mangostin transethosome gel with values respectively 11.1584%; 13.4918%; 12.3556% and the three preparations follow the three compartment model or lag model and the significance value obtained is p <0.05. Based on the results of the study, it can be concluded that the transethosome gel formula has good characteristics according to the range of test parameter requirements and has the potential as a wound healing gel.
