Abstract
Alpha-mangostin as the main xanthone compound in mangosteen peel (Garcinia mangostana L.), exhibits pharmacological activities, including anti-inflammatory, making it a potential candidate for topical formulations. However, due to its limited penetration, a transethosome based patch is needed to optimize its effectiveness and usability. This study aims to develop a transethosomal patch preparation of alpha mangostin by evaluating the physicochemical characteristics to obtain the best formula. Transethosome patch were made by solvent casting method in 3 formulas that have different concentrations of propylene glycol. The best formula results were obtained in formula 3 with concentration of propylene glycol 20%, this can be seen from the characterization results including organoleptic, thickness, folding power, weight uniformity, % elongation, moisture, pH, and swelling index. The organoleptic results have an appearance, yellow in color, smooth surface, elastic, and have a distinctive odor of extract. The evaluation results of thickness, folding power, weight uniformity, elongation, pH, moisture, swelling index were respectively 0.300 ± 0.01 mm; >300 folds; 0.854 ± 0.027 mg; 104.875 ± 0.28%; 7.15 ± 0.03; 3.077 ± 0.01%; and 249.285 ± 2.899%. Compartmental analysis using WinSAAM® software showed that the patch followed the lag compartment model with 0th order release kinetics and the release mechanism followed Korsmeyer-Peppas. Stability test using the cycling test method on the best formula of transethosome patch showed no significant changes in organoleptic, content, moisture, and pH in the preparation with a value of p>0.05. Observation of content uniformity showed uniform results with % recovery 59.758% ± 1.266 and the morphology of the transethosome was still visible with a spherical vesicle shape in the patch preparation. The research results show that the transethosome patch of alpha-mangostin in formula III has good characteristics, stability, and penetration while maintaining the morphology of transethosomes, making it suitable for development as a transethosome patch.
