Abstract
Coffee is a plantation commodity that has high commercial value and is important for the Indonesian economy. However, the problem often faced by farmers is the difficulty of obtaining coffee seeds with superior quality. The solution that can be done is to carry out innovative new techniques through propagation with tissue culture techniques. The main challenge in in vitro propagation of coffee plants is the high level of contamination in explants that can inhibit growth. This study was conducted to determine the solution concentration and immersion time in sodium hypochlorite (NaOCl) that is effective for sterilizing coffee leaf explants. The research was conducted at the Tissue Culture Laboratory, Faculty of Agriculture, Sriwijaya University from December 2024 to January 2025. Observations in this study used observation, quantitative, numbering, and descriptive methods. This study used five sterilants consisting of liquid Detergent, Streptomycin sulfate, Benomyl, Alcohol, and NaOCl with different concentrations and length of immersion in NaOCl. Each treatment in this study was repeated 3 times and each treatment consisted of 10 culture bottles so that 150 experimental units were obtained. The results showed that (P5), namely explants sterilized using a treatment consisting of (Liquid Detergent 5 minutes, Streptomycin Sulfate 10 minutes, Benomyl 10 minutes, 70% Alcohol 3 seconds, NaOCl 0.75% 3 minutes, and NaOCl 1.25% 3 minutes) produced the highest percentage of live explants at 90% on the 31st day after inoculation. In addition, this treatment also succeeded in reducing the contamination up 10%.
