Abstract

Green betel leaves (Piper betle L.) contain flavonoids, tannins, and saponins that have the potential as natural hemostatic agents. This study aims to determine the effect of ethanol extract of green betel leaves on bleeding time and blood clotting time in male mice (Mus musculus), and to determine its effective dose. This study was divided into five treatment groups, namely negative control (1% NaCMC), positive control (500 mg/kgBW tranexamic acid), and ethanol extract of green betel leaves with doses of 100 mg/kgBW, 200 mg/kgBW, and 300 mg/kgBW. Bleeding time was observed using the Duke method and blood clotting time using the slide method. The results showed that green betel leaf extract was able to accelerate hemostasis time significantly (p <0.05), with a dose of 200 mg/kgBW showing the most optimal effectiveness and approaching the results shown by the positive control. The hemostasis mechanism of this extract is thought to be related to platelet activation by flavonoids, as well as the role of tannins and saponins in vasoconstriction and blood protein binding. Based on these results, the ethanol extract of green betel leaves has the potential as a natural hemostatic agent.
