Abstract
The purposes of this research were to determine the effect best method of extraction in determining the fatty acid profile and to know the profile of amino acids contained in Mudskipper fish (Boleophthalmus sculptus). This research was conducted from August until December 2016 by using laboratory experimental methods and data was analysed descriptively. Analysis of the fatty acid profile of the Mudskipper fish used GC (Gas Chromatography) and amino acid analysis used a UPLC (Ultra Performance Liquid Chromatography), except for triptofan used a HPLC (High Performance Liquid Chromatography). Fatty acid profile data obtained were statistically analyzed using independent sample t-test test. The analysis showed that the Mudskipper fish extracted with different extraction methods produce fat content varies by sequential numbers, fat content in the extraction with hexane average of 1.8260% while the fat content in the extraction with chloroform:methanol 2:1 v/v on average of 2.6163. The content of fatty acids identified in mudskipper fish extracted with hexane as many as 19 types of fatty acids which consists of 10 types of saturated fatty acids and 9 types of unsaturated fatty acids while the fatty acids extracted with chloroform:methanol 2:1 v/v as many as 24 different types of fatty acids which consists of 11 kinds of saturated fatty acids and 13 kinds of unsaturated fatty acids. On the test of independent sample t-test can be concluded that there is no difference in the average amount of fat, saturated fatty acids, unsaturated fatty acids, omega-3, omega-6, omega-9, AA, EPA and DHA between hexane extraction and extraction of chloroform:methanol 2:1 v/v. Analysis of amino acids in Mudskipper fish produce a complete amino acid that is 18 amino acids which consists of 10 kinds of essential amino acids and 8 kinds of non essential amino acids.
