Abstract
Diuretics are used as drugs to treat hypertension. Diuretics work to remove excess fluid in the body by increasing the process of urine formation. The secondary metabolite content in jackfruit leaves which is thought to have potential as a diuretic is phenolic compounds. This study aims to determine the diuretic activity of the ethyl acetate fraction of jackfruit leaves. Total phenolic content was determined by colorimetric method. Diuretic activity testing was carried out on 6 groups of test animals including normal, negative, positive groups, and three groups of test doses of ethyl acetate fraction 25, 50 and 100mg/200gBW. The total phenolic content of the ethyl acetate fraction of jackfruit leaves was 51 mg GAE/g. The results showed that the 100 mg/200gBW dose group could increase the excretion of urine volume and the Na+, K+ and Clions were higher than the positive group (p<0.05). The dose of ethyl acetate fraction of jackfruit leaves at a dose of 100mg/200gBW was the most effective as a diuretic compared to doses of 25 mg/200gBW and 50 mg/200gBW. The higher the dose fraction, the more effective it is as a diuretic. This is indicated by the increase in the volume of urine output and the levels of Na+, K+ and Clions in the test animals. The effective dose (ED50) of the jackfruit leafethyl acetate fraction was 27.485 mg/200gBW. Diuretik bekerja mengeluarkan cairan berlebih dalam tubuh dengan meningkatkan proses pembentukan urin. Kandungan metabolit sekunder pada daun nangka yang diduga berpotensi sebagai diuretik yaitu senyawa fenolik. Penelitian ini bertujuan untuk mengetahui aktivitas diuretik dari fraksi etil asetat daun nangka. Kadar fenolik total ditentukan dengan metode kolorimetri. Pengujian aktivitas diuretik dilakukan terhadap 6 kelompok hewan uji meliputi kelompok normal, negatif, positif, dan tiga kelompok uji dosis fraksi etil asetat 25, 50 serta 100mg/200gBB. Kadar fenolik total dari fraksi etil asetat daun nangka sebesar 51 mg GAE/g. Hasil penelitian menunjukkan bahwa kelompok dosis 100 mg/200gBB dapat meningkatkan pengeluaran jumlah volume urin dan ion Na+, K+ serta Cllebih tinggi dibandingkan kelompok positif (p<0,05). Pemberian dosis fraksi etil asetat daun nangka dosis 100 mg/200gBB dan 50 mg/200gBB. Semakin tinggi dosis fraksi maka semakin efektif sebagai diuretik. Hal ini ditunjukkan dari peningkatan pengeluaran jumlah volume urin dan kadar ion Na+, K+ serta Clpada hewan uji. Dosis efektif (ED50) dari fraksi etil asetat daun nangka sebesar 27,485mg/200gBB.
