Abstract

Suatu penelitian untuk mempelajari aktivitas antibakteri kompleks kitosan monosakarida pada matriks pangan surimi ikan gabus. Penelitian ini dilaksanakan pada bulan April sampai Juni 2014 di Laboratorium Teknologi Hasil Perikanan dan Laboratorium Budidaya Perairan Universitas Sriwijaya. Penelitian ini menggunakan rancangan acak kelompok. Perlakuan kompleks kitosan monosakarida terdiri dari larutan kitosan (sebagai kontrol), kompleks kitosan glukosa, kompleks kitosan galaktosa dan kompleks kitosan fruktosa. Parameter yang diamati meliputi analisis produk reaksi Maillard (Maillard reaction productsfMKPs), analisis kimia dan analisis aktivitas antibakteri terhadap tiga bakteri pada hasil perikanan (Bacillus subtilis, Listeria monocytogenes dan Vibrio cholerae) pada matriks pangan dengan metode total plate count/TPC dan metode kertas cakram (in vitro). Perlakuan dalam penelitian berpengaruh nyata terhadap analisis MRPs dan analisis TPC surimi, tetapi tidak berpengaruh nyata terhadap diameter daerah hambatan (DDH) bakteri. Analisis MRPs menghasilkan absorbansi yang berkisar antara 0,052-0,248, analisis metode cakram menghasilkan DDH terhadap B subtilis yang berkisar antara 10,5-11,75 mm, L. monocytogenes berkisar antara 10-11,5 mm dan V cholerae berkisar antara 9,25-10,75 mm. Analisis TPC menunjukkan kompleks kitosan monosakarida lebih efektif menghambat bakteri Gram negatif (V cholerae) daripada Gram positif (B. subtilis dan L. monocytogenes). Kompleks kitosan monosakarida terbukti tetap memiliki aktivitas antibakteri meskipun terjadi interaksi dengan matriks pangan.
