Abstract

The kipahit plant (Tithonia diversifolia) belongs to the Asteraceae family originating from South and North America which has bioactivity as anti-inflammatory, anti- fumigant, anti-viral, anti-diabetic and antibacterial. The antibacterial activity of extracts, fractions and isolated compounds against pathogenic bacteria that cause skin diseases such as Staphylococcus epidermidis and Streptococcus pyogenes has never been reported. Isolation of secondary metabolites begins with extraction using the maceration method with methanol solvent followed by the liquid-liquid fractionation process using n-hexane: ethyl acetate respectively. Separation and purification of the isolated compounds was continued by vacuum liquid chromatography (VLC) and radial chromatography using a chromatotron. The isolated compound is a yellow solid (26.2 mg). Based on the analysis of UV, IR and NMR spectra (1H-NMR, 13C- NMR, HSQC and HMBC), the isolated compound was a polyketide group namely 6-acetyl-7- hydroxy-2,2-dimethylchromen which was supported by the references. Antibacterial activity test on methanol extract, n-hexane fraction and ethyl acetate fraction with various concentrations from 6.250 – 50.000 ppm and compounds 6-acetyl-7-hydroxy-2,2-dimethylchromen with various concentrations from 500 – 4000 ppm have inhibitory activity against test bacteria with categories weak which was indicated by the percentage value of the inhibition zone which is < 50%.
