Abstract
Oil palm empty fruit bunches (EFBF) waste is a waste that takes a long time to degrade, this is due to the high cellulose content in EFB waste. Cellulose is a hydrolyzed polysaccharide to produce cellobiose and glucose monomers. Cellulase can be produced by plants, yeast, bacteria and cellulolytic fungi. Fungi have the ability to decompose cellulose found in dead plant tissues into simpler compounds due to their ability to produce lignocellulolytic enzymes. Cellulolytic fungi are microorganisms capable of degrading cellulose by producing cellulase enzymes. Cellulase is an enzyme complex that plays a role in the hydrolysis of cellulose consisting of endoglucanase, ecoglucanase, and cellobiase. This study aims to determine the genus of fungi that have high cellulase activity values isolated from OPEFB waste. The research was conducted from January to May 2023, taking place at the Microbiology Laboratory, as well as the Genetics and Biotechnology Laboratory, Department of Biology, FMIPA, Sriwijaya University, Indralaya. The stages of the research were isolation and purification of fungi, qualitative selection of cellulolytic function, preparation of crude extracts of cellulase enzymes, preparation of glucose standard curves, quantitative testing of cellulase enzyme activity using the DNS method, characterization and identification of high cellulase activity values. The results showed that 8 out of 11 fungi isolates had the ability to produce cellulase enzymes. Isolates TS2B1, TS1L1, TS3L2 and TS3L3 had high cellulase activity values respectively 5.7 (U/ml), 6.5 (U/ml), 6.1 (U/ml), 5.8 (U/ ml) based on cellulase enzyme activity test with DNS method. Isolates TS2B1, TS1L1, TS3L2 and TS3L3 had high cellulase activity values respectively 5.7 (U/ml), 6.5 (U/ml), 6.1 (U/ml), 5.8 (U/ ml) based on cellulase enzyme activity test with DNS method. Isolate TS2B1 was identified as the genus Penicillium, isolates TS1L1 and TS3L3 were identified as the genus Fusarium and isolate TS3L2 was identified as the genus Trichoderma
