Abstract

Ibu perokok pasif dapat terpapar nikotin dalam darah yang dihasilkan dari pembakaran rokok yang disebabkan oleh suami perokok. Paparan nikotin pada ibu perokok pasif dapat menyebabkan masalah kesehatan, seperti asma, serangan jantung, gangguan kehamilan hingga kematian. Dosis maksimum nikotin dalam tubuh manusia adalah 60 mg/mL. Penelitian ini bertujuan untuk memvalidasi metode dan penentuan kadar nikotin dalam darah ibu perokok pasif menggunakan Gas Chromatography-Mass Spectrometry (GC-MS), dengan parameter analisis, seperti linieritas kurva kalibrasi, limit deteksi (LOD), danlimit kuantisasi (LOQ). Sampel diambil masing-masing 3 mL dari darah 5 ibu perokok pasif dengan kode sampel A-E. Selanjutnya darah dipisahkan dan diekstraksi menggunakanpelarut isoamil alkohol. Hasil penelitian menunjukkan bahwa kurva linier dari rentang konsentrasi 0,01-1,00 mg/L dengan persamaan regresi linier y = 9250,3x + 82957 dengan nilai koefisien korelasi (R2) = 0,9958, nilai LOD sebesar 0,018 mg/L dan nilai LOQ sebesar 0,074 mg/L. Kadar nikotin dalam sampel plasma darah A-D berkisar dari 0,07-0,5 mg/L
