Abstract
The main bacteria that plays an important role in the biofilm formation is Streptococcus mutans. Aloe vera has demonstrated antibiofilm activity against S. mutans and can be made into a jelly candy product. This study aims to determine the antibiofilm activity of aloe vera jelly candy against S. mutans. Method: This study used an in vitro experimental laboratory. The antibiofilm activity against S. mutans was determined using microtiter plate method (using 96 wells) on four sample groups: aloe vera jelly candy, jelly candy without aloe vera, negative control (S. mutans suspension) and media control (Brain Heart Infusion Broth). Antibiofilm activity was calculated by measuring absorbance or optical density value using a microplate reader at a wavelength 450 nm, then calculating the average of absorbance value. Data were analyzed using One Way ANOVA test. Results: One way ANOVA test showed that there was a significant difference average of absorbance value between all groups (p<0,05). Aloe vera jelly candy was able to significantly reduce the absorbance value compared to jelly candy without aloe vera and the negative control, but was not able to compete with media control (p<0.05).Conclusion: Aloe vera jelly candy has antibiofilm activity against S. mutans, therefore this jelly candy has the potential to prevent caries. Keywords: antibiofilm, aloe vera, jelly candy, Streptococcus mutans. Bakteri utama yang berperan penting dalam proses pembentukan biofilm adalah Streptococcus mutans. Lidah buaya terbukti memiliki aktivitas antibiofilm terhadap S. mutans dan dapat dijadikan produk dalam bentuk permen jeli. Penelitian ini bertujuan untuk mengetahui aktivitas antibiofilm permen jeli lidah buaya terhadap S. mutans. Metode: Penelitian ini menggunakan studi eksperimental laboratoris secara in vitro. Uji antibiofilm S. mutans menggunakan metode microtiter plate (menggunakan well 96) pada empat kelompok sampel yaitu permen jeli lidah buaya, permen jeli tanpa lidah buaya, kontrol negatif (suspensi S. mutans) dan kontrol media (Brain Heart Infusion Broth). Aktivitas antibiofilm dihitung dengan mengukur nilai absorbansi atau optical density menggunakan microplate reader pada panjang gelombang 450 nm, kemudian dilakukan perhitungan rata-rata nilai absorbansi. Data dianalisis menggunakan One Way ANOVA test. Hasil: Hasil uji one way ANOVA menunjukkan terdapat perbedaan rerata nilai absorbansi yang signifikan antara semua kelompok (p<0,05). Permen jeli lidah buaya mampu menurunkan nilai absorbansi secara signifikan dibandingkan dengan permen jeli tanpa lidah buaya dan kontrol negatif, namun belum cukup baik dibandingkan kontrol media (p<0,05). Kesimpulan: Permen jeli lidah buaya memiliki aktivitas antibiofilm terhadap S. mutans, sehingga permen jeli ini berpotensi untuk mencegah karies.
