Abstract
Sambiloto leaves (Andrographis paniculate (Burm. F) Wall ex. Ness) contain andrografolid and flavonoids as the main compounds that have potential as antibacterial. Nanoemulsion with a combination of tween 20 and PEG-400 was carried out to produce preparations that have better antibacterial effects and are thermodynamically stable. This study was conducted to determine the characteristics and stability of nanoemulsion preparations using the accelerated test method according to the ICH Q1A (R2) standard and to test antibacterial activity against Propionibacterium acnes bacteria using the disc diffusion method to see the diameter of the inhibition zone and liquid dilution to determine the Minimum Inhibitory Concentration (KHM). The characteristics of nanoemulsion of sambiloto leaf extract showed a transparent green color with a distinctive aroma and liquid texture and a pH value of 5.483 ± 0.004; viscosity of 1.010 ± 0.021 cP; specific gravity 1.027±0.001gr/cm3 ; percent transmittance 99.466±0.047%; absorption efficiency 96.224±0.001%; droplet size 162.220±73.773nm; polydispersity index 0.477±0.153; and zeta potential -23.966±1.862. Based on the results of the stability test, the shelf life of the nanoemulsion preparation calculated based on first-order kinetics at 25o C for 25.9901 days and at 5o C for 27.1137 days. Nanoemulsion of sambiloto leaf extract showed a strong inhibitory response with a diameter of 12.333 ± 1.247 mm and a minimum inhibitory concentration (KHM) at a concentration of 40 mg/ml. Based on the results obtained, it can be concluded that nanoemulsion preparations have good characteristics and stability as well as strong antibacterial activity.
