Abstract
The batik industry has an important role in the Indonesian economy, but the large number of batik industries in Indonesia is not matched by proper wastewater treatment. One of the components of batik wastewater is the synthetic dye Naphthol ASBO-Blue B. Biological treatment of dye waste with bacteria as a biodecolorization agent needs to be done to reduce the negative effects caused by dye waste. This study aims to obtain thermophilic bacterial isolates that have the potential to decolorize Naphthol ASBO-Blue B dye, obtain thermophilic bacterial species that have the potential as biodecolorization agents based on 16S rRNA gene analysis, and determine the decolorization mechanism by analyzing the chromatogram pattern of biodecolorization products. Six bacterial isolates from Tangkuban Perahu hot springs were able to decolorize Naphthol ASBO-Blue B. at a concentration of 100 ppm for 7 days with the largest percentage of biodecolorization ability owned by isolate DS2D which is 53.03 ± 0.28%. The results of the KLT product analysis after biodecolorization by isolate DS2D, indicated the breaking of the azo bond so that the degradation of the dye occurred. Isolate DS2D has a streptobacil shape, is gram-positive, and produces endospores. The DS2D isolate sequence showed high similarity (97.90%) with Bacillus tropicus strain MCCC 1A01406, and formed a monophyletic clade on the phylogenetic tree.
