Abstract

High doses of antibiotics use continously can increase an antibiotic resistance, therefor a research for new antibiotics is needed to reduce the possibility of resistance. Actinomycetes are one of the microorganisms that have commercial value because of their ability to produce antibiotics. 80 % of antibiotics on the market today are produced by Streptomyces. Exploration of Actinomycetes from swampland revealed 19 isolates and 4 isolates of which ACT 8, ACT 10, ACT 11 and ACT 13 had high antibacterial activity against Escherichia coli and Staphylococcus aureus based on morphological and biochemical characterization of the four isolates suspected to be genus Streptomyces. This research was conducted from August 2021 to March 2022 in Laboratory of Microbiology, Laboratory Genetic and Biotechnology, Biology Departmen, Faculty of Mathematics and Natural Science, Sriwijaya University. This research was conducted to obtain Streptomyces spp that have the potential to produce antibacterial agains E.coli dan S.aureus. The stages of the research carried out included confirmation test of Streptomyces spp isolates antibacterial ability, cultivation and extraction, antibacterial activity test, minimum inhibitory concentration of Streptomyces spp extract, thin layer chromatography (TLC) an bioautography tests. The conclusion obtained from this study is that the values of the Minimum Inhibitory Concentrations (MIC) for Streptomyces ACT 8, Streptomyces ACT 10, Streptomyces ACT 11 and Streptomyces ACT 13 isolates against E. coli were 15 g/mL, 31.25 g/mL, 15 g, respectively. /mL and 5 g/mL, while the test bacteria for S. aureus were 31.25 g/mL, 31.25 g/mL, 62.5 g/mL and 10 g/mL, respectively. The active compound isolated from Streptomyces spp. The active compounds isolated from Streptomyces ACT 8, Streptomyces ACT 11 and Streptomyces ACT 13 that were able to inhibit Escherichia coli were flavonoids, terpenoids and phenols, while the active compounds of Streptomyces ACT 10 were flavonoids and phenols. The active compound of the secondary metabolite extract of Streptomyces spp. which are able to inhibit Staphylococcus aureus are flavonoids, phenols and terpenoids from StreptomycesACT 13; flavonoids from Streptomyces ACT 8; terpenoids and phenols from Streptomyces ACT 10; Streptomyces ACT 11 flavonoids and phenols.
