Abstract
Bacteria in symbiosis with sponges are potential candidates in the field of biotechnology because they can produce bioactive compounds such as antioxidants, antidiabetic, antibacterial, anticancer, and antibiofilm. Identification is carried out molecularly using 16S rRNA gene analysis because it can see the similarity of each bacterial species with close to 99% similarity. This research aims to isolate and determine the type of sponge symbiont bacteria originating from the aquatic ecosystem of Enggano Island. The research was conducted at the National Research and Innovation Agency (BRIN) Laboratory, Cibinong, Bogor, West Java, in September-December 2023. Sample processing began with isolation preparation, multilevel dilution, bacterial isolation/ purification, gram staining, macromorphological observation, DNA extraction genome, quantitative testing, DNA amplification (PCR), electrophoresis, sequencing, and data analysis. Based on the research results, 50 isolates were isolated from the Stylissa massa sponge using SWC, MA, ISP-2, and Zobell 2216 media. 27 isolates were gram-positive bacteria and the other 23 isolates were gram-negative bacteria. Ten isolates that showed the best gram-positive (6 isolates) and gram-negative (4 isolates) pigments were selected for identification. The identification results showed that isolate SM3 had genetic similarities to Acinetobacter baumannii, SM10 to Brevibacterium casei, SM11 and SM12 to Pseudovibrio denitrificans, SM17 to Kocuria variance, SM18 to Psychrobacter piechaudii, SM22 and SM23 to Staphylococcus epidermidis, SM14 and SM25 to Staphylococcus hominis. This type of bacteria has great potential in the field of biotechnology.
