Abstract

Temu ireng (Curcuma aeruginosa Roxb.) is one of the medicinal plants that has been reported to have antioxidant, antibacterial, anti-inflammatory, and hepatoprotector activities, but this plant has not been widely studied regarding its toxicity. This study conducted an acute toxicity test of ethanol extract of temu ireng rhizome against female Wistar strain rats using the Acute Toxic Class method. The results of standardization of ethanol extracts of temu ireng rhizomes show thick, dark brown extracts and have a distinctive odor of extracts, water soluble juice content 43.33 ± 5.77; ethanol soluble juice content 83.33 ± 11.54; moisture content 6.67 ± 1.15; total ash content 12.33 ± 0.76; acid insoluble ash content 0.51 ± 0.28 and specific gravity 1.03 ± 0.01 (mg/dL). Toxicity observations showed that death and toxic symptoms occurred in the test animals. There was no change in body weight of rats between before and after administration of the extract (p>0.05). In the examination of biochemical parameters, there were no significant differences (p>0.05) between the normal and treatment groups, while the levels of SGOT and SGPT had significant differences (p<0.05). Biochemical levels at a dose of 10,000 mg/kgBB were SGOT 146.06 ± 13.50 U/L, SGPT 72.58 ± 1.8 U/L, creatinine 0.44 ± 0.06 mg/dL, ureum 29.28 ± 0.78 mg/dL, LDL 5.67 ± 0.59 mg/dL and triglycerides 78.83 ± 12.4 mg/dL. Based on the results of the study, it can be concluded that the temu ireng rhizome extract meets the standardization requirements, the LD50 value of temu ireng rhizome extract is 10,000 mg / kgBB, Giving ethanol extract of temu ireng rhizome affects the liver, kidney and heart organs macroscopically and microscopically.
